Dean/Jo:

Jo Harvelle and Dean Winchester met under interesting circumstances. After the death of their father, Sam and Dean drove off to the Roadhouse after cracking the voicemail code on John's cellphone and listening to an old message from Ellen, Jo's mother. When they arrived, both boys were met with loaded guns to their backs, Jo holding a shotgun to Dean's and Ellen holding a handgun to Sam's.

The flirting started immediately, as is to be expected from Dean Winchester. He and Jo never actually hooked up, however, since it was the "wrong place, wrong time." Not even when she defied her mother and the brothers' orders by going on a hunt with them and serving as bait did anything happen, but Dean's protective instinct was dumped onto Jo as well as his little brother. When the three returned to the Roadhouse, all a little worse for the wear, Ellen informed Jo that her father had been killed because of John.

When Jo told Dean this information, he was shocked. They didn't speak again for quite a long time until Sam was possessed by a demon and that demon, in an attempt to make Dean kill his brother, tracked down Jo at her new place of work (she had left the Roadhouse shortly after learning about her father's death) and held her hostage. Dean promised to call her. So far, he hasn't.
